I think I've worked at least part of it out. I can open hyperlinks in several fields (I've tried Link To PDF, URL, and Notes), and pressing Ctrl+G (or Reference/Open Link) opens whatever is in the URL field (and it works from both the library and the reference window).
There are at least three issues.
First, as has already been noted, spaces need to be entered as %20. Other characters, such as #, also need to be entered with the relevant http code (# is %23). The link needs to be prefixed with http:// or file:// Using the Link To... command automatically gets it right. In fact, the number of forward slashes after file: seems arbitrary: I've had success with none, two, and three.
Second, as I noted several months ago, you can't have two entries in the URL field. Unlike other fields, you can't just press enter and type a new link. Endnote seems to understand that they are different entities (words on intervening lines appear black, and the pointer is an I-beam rather than a pointing finger), but the error message when you try to open the link says "unable to open link <link 1><any intervening text><link 2><etc.>"
So, keep your URL fields to one entry (whichever one you want to open by default), and place all others in a different field... notes, for example, or link to PDF. Ironically, my URLs tend to go in the link to PDF field, and my local copies of PDFs go in the URL field.
THIRD, AND MOST IMPORTANTLY, Endnote does not always apply changes to the URL field until you close the record window. If you make a change to the URL field, press Ctrl+G, and still get an error, try closing the record and opening it again. The exception is when you use the Link To... command to enter text into the URL field, in which case it seems to update straight away.
